WP1 Expression profiling by microarray and microCAGE of chordate dopaminergic neurons

Description of Work
This work package aims to obtain high quality expression profiling data from single dopaminergic neuron cells isolated from mouse, zebrafish and Ciona. Expression profiles will be generated from microarray analysis of quality-ensured RNA. The CAGE (cap-analysis gene expression) library will also be prepared as part of this work package.

WP2 Screening of chordate cis-regulatory elements

Description of Work
The cis-regulatory elements of Ciona will be subjected to medium throughput analysis while those from the zebrafish will be analysed using high throughput technologies. The interactions of cis-regulatory elements in zebrafish embryos will also be investigated.

WP3 Data Analysis and Network Reconstruction

Description of Work
The aim of this workpackage is to reconstruct the network of dopaminergic neurons for each of the four organisms under study and compare them. A comparative network analysis of different sub-types of neurons will be completed simultaneously. In conjunction with microarray analysis, micro-CAGE will be employed to identify core promoters and their usage as well as non-coding RNA and their potential networks and finally to predict cis-regulatory elements in selected chordate genes.

WP4 Network perturbations

Description of Work
The main objective of this workpackage is to identify potential therapeutic targets. Expression profiling of responses to experimental perturbations (gain/loss of function) will be used to infer Cionas epistatic relationship and network position. Inference of zebrafish transcription factors and ncRNAs will also be undertaken.

WP5 Training, Dissemination and exploitation

Description of Work
Several initiatives to disseminate the progress and results of Dopaminet will be undertaken. The primary source for information and discussion of the project will be the website which will be updated a minimum of every six months with information on related European projects also being noted there. Every six months dissemination material will be prepared and dispersed as widely as possible. On the training side, the project will hold a summer school each year for the duration of the project.

WP6 Management and coordination

Description of Work
Management of the project will include such areas as controlling the finances and ensuring the meeting of deadlines. The management team will also be responsible for organising and implementing project meetings.
